1. The cells need to be re-adapted due to the change of different culture medium or serum.
2. The reagents are improperly stored, and some necessary components for cell growth such as glutamine or growth factors in the culture medium are exhausted or lacking or have been destroyed.
3. There is a small amount of bacterial or fungal contamination in the culture in the cell shaker.
4. The initial concentration of inoculated cells is too low.
5. Cells have aged.
6. Mycoplasma contamination
Suggested solution:
1. Compare the composition of the new medium and the original medium, and compare the new serum and the old serum to support cell growth experiments. Allow cells to gradually adapt to the new medium.
2. Change into freshly prepared culture medium, or add glutamine and growth factors.
3. Incubate with antibiotic-free medium and replace the culture if contamination is found. The culture medium should be stored at 2-8°C in the dark. Serum-containing complete medium is stored at 2-8°C and used up within 2 weeks.
4. Increase the starting concentration of inoculated cells.
5. Replace with new seeded cells.
6. Isolate the culture and detect mycoplasma. Clean the stand and incubator. If mycoplasma contamination is found, replace with a new culture.
