Cause analysis of precipitate in cell culture flask—temperature

Cell culture is a method for cells to survive, grow, reproduce and maintain their main structures and functions by mimicking the environment in vivo in vitro. Cell culture bottle  is a kind of cell consumable commonly used in adherent cell culture. In the process of cell culture, we often find some impurities accumulation in the liquid. There are many reasons for this situation, and temperature is also one of the common reasons.
95The presence of precipitation in the cell culture flask may be the result of cell contamination. If contamination is excluded, turbidity in the cell culture medium is usually interpreted as precipitation of metal elements, proteins, and other medium components. Most precipitates impair normal cell proliferation because they alter the composition of the medium by chelating nutrients and other required components. The precipitate may be observed microscopically and may interfere with experiments requiring imaging analysis.
 
In cell culture, temperature is one of the main factors causing precipitation. When the temperature changes extremely, high molecular weight plasma proteins will be precipitated from the solution. Heat inactivation and freeze-thaw cycle can promote protein degradation and precipitation. Because the liquid or reconstituted medium is kept in cold storage between uses, salt may settle, especially in 10X or other concentrated storage solutions.
 
Of course, precipitation appears in the cell culture bottle. If it is determined that the temperature is the cause, attention should be paid to the storage environment and operation method of the culture medium to avoid repeated freezing and thawing, which can reduce the probability of precipitation.
